The Axor Biosystems microplate Electrophoresis Strips are made up of 2, 4, or 8 independent Electrophoresis Cells. Each cell contains 3-dimensional structures with fluidic chambers, controlling ports, and fluidic channels. The conventional 2-dimensional slab gel is transformed into a compact structure as small as 9 mm x 9 mm, without significantly impacting the resolution.
Optimized buffers, composite separation matrices, and disposable electrodes that minimize hydrolytic gas generation and reduce pH buffer changes are placed within the cell. The designs facilitate gas escape and provide buffer exchange capability. A top cover switches power to the electrodes placed within each cell.
Sample is pre-concentrated by applying a voltage across an input electrode and a concentration electrode, resulting in the analyte coalescing onto the concentration electrode - useful for dilute samples.
The voltage is then switched between the input electrode and an electrode placed at the end terminus of the separation channel - resulting in the migration of the materials through the channel. A camera array monitors the migration for the electrophoresis microplate in real-time. The ML-based software calculates the Molecular Weights of the migrating materials based on non-interfering fiducials loaded with the sample.
When the required Molecular Weight band reaches a purification junction, the voltage is now applied across the purification channels resulting in the required band migrating through the purification ports and into a purification well.
A second fragment can be isolated by applying the voltage to the input well and a second purification well resulting in an additional fragment migrating into the second purification well.
As each electrophoresis cell is isolated from every other cell in the strip, a 500 bp band can be isolated in one cell and and a 1000 bp band in another cell, for example. An additional advantage is that strips supporting different applications can be run together, e.g., a protein analysis strip and a DNA purification strip can be processed together.
