The sample sheet template shown below is available with purchase of the 1 Box System.
Select User ID (predefined by the Admin.) from the pull-down menu.
Input Sample IDs by typing in the values, by using a barcode reader or importing from a table.
Input Sample Name, if needed, by typing in the values or importing from a table.
Select Sample Type from the pull-down menu [DNA, RNA, Protein, Cells].
Select Sample volume (in uL) from the pull-down menu [<10uL, 10-100uL].
Select Method to be used from the pull-down menu [Preparative, Analytical, Capillary].
Select Purification Process from the pull-down menu [Extract, Subtract, Not Applicable]. This column will be grayed out if ‘Preparative’ was not selected in the Method column.
Input Molecular Weights and the range of M.W. for the first ‘Extraction’ or ‘Removal’. These columns will be grayed out if Preparative’ was not selected in the Method column.
Input Molecular Weights and the range of M.W. for a second extraction, if required. These cells will be grayed out if Preparative’ was not selected in the Method column.
The sheet will calculate the optimal strips required to perform the electrophoresis application. Select a strip from the displayed pull-down menu of Recommended Strips.
Remove the sample-sheet recommended strips from the Axor kits. Place in the included strip holder.
Enter the Strip ID of selected strip(s) into the Sample Sheet. Save sheet for importing into the system software later.
Arrange the samples in the same order as the strips in the strip holder - to simplify loading samples.
Add ‘10x Sample Buffer’ (present in the kit) to each sample. Mix well. Centrifuge briefly. Note: The Sample Buffer is specific to a kit. Do not use a Sample Buffer from one kit with strips from another kit.
Pipette the mixed samples into the Sample Loading Well (for < 10 uL) or the Input Well (10-300uL) depending on the strip format. The sample wells are marked 's' in each electrophoresis cell. The input wells are marked 'i'. A standard pipette tip will pierce through the protective layer covering the well. Please, see the detailed instructions associated with each kit for more information.
Start the Axor 1 Box app and login with username.
Click Import Sample Sheet, navigate to where the sheet is stored and select. The sheet will be loaded into the software.
Click on Open Drawer. Place the prepared strips into the drawer. Click on Close Drawer. The IDs on the strips will be read to confirm match with the sample sheet.
Click on Start Run to initiate the run.
Data is collected in real-time and displayed for the selected cell. Click on a row in the sample sheet or a cell in the plate map to preview Results for that cell.
Results can be exported on Run completion by clicking on Export.
On Run Complete, the purified materials are retained in the Wells by a low voltage till you click on Open Drawer button. Remove the strip holder when the drawer opens.
Pipette out the purified samples from the Purification Wells (for ‘Extract’) or the Output Well (for ‘Subtract’) within 10 minutes.
